Intracellular DNA sensing by neutrophils and amplification of the innate immune response

نویسندگان

چکیده

As the first responders, neutrophils lead innate immune response to infectious pathogens and inflammation inducing agents. The well-established pathogen neutralizing strategies employed by are phagocytosis, action of microbicide granules, production ROS, secretion neutrophil extracellular traps (NETs). Only recently, ability sense respond pathogen-associated molecular patterns is being appreciated. This review brings together current information about intracellular recognition DNA proposes models signal amplification in response. Finally, clinical relevance sensing non-infectious diseases including malignancy also discussed.

Neutrophils in the innate immune response.

Polymorphonuclear leukocytes (PMNs or neutrophils) are an essential component of the human innate immune system. Circulating neutrophils are rapidly recruited to sites of infection by host- and/or pathogen-derived components, which also prime these host cells for enhanced microbicidal activity. PMNs bind and ingest microorganisms by a process known as phagocytosis, which typically triggers prod...

Innate Immune Sensing of DNA

When a pathogen attacks, the immune system rapidly mobilizes host defenses in order to reduce the microbial burden and limit damage to the host [1]. Innate immunity is the first line of defense and relies on germ line–encoded pattern recognition receptors (PRRs) such as the Toll-like receptors (TLRs), which sense microbial products that are not normally found on or in mammalian cells. The consi...

Innate Immune DNA-Sensing Pathways

Recognition of foreign DNA in intracellular compartments or in the cytoplasm sounds an alarm bell signaling pathogenic invasion. In response, innate immune DNA-sensing pathways launch an antimicrobial type I interferon (IFN) response, which becomes damaging to the host if unregulated or inappropriately activated by self-DNA. This Select highlights recent insights into activation and regulation ...

Innate Immune Response in Implant-Associated Infections: Neutrophils against Biofilms

Biofilm has been recognized as a well-protected form of living for bacteria, contributing to bacterial pathogenicity, particularly for opportunistic species. Biofilm-associated infections are marked by their persistence. Extensive research has been devoted to the formation and composition of biofilms. The immune response against biofilms remains rather unexplored, but there is the notion that b...

Tamoxifen Augments the Innate Immune Function of Neutrophils Through Modulation of Intracellular Ceramide

Tamoxifen is a selective oestrogen receptor modulator widely used for the treatment of breast cancer. In addition to its activity as an oestrogen receptor agonist/antagonist, tamoxifen also modulates sphingolipid biosynthesis, which has been shown to play an important role in the regulation of neutrophil activity. Here, we find that tamoxifen stimulation enhances several pro-inflammatory pathwa...
